The required technique to control the actual separation temperature is often underestimated and, in many cases, not well understood in UHPLC. Thermostatting has to be effective, but must not compromise the achievable column efficiency. On the other hand, it is very important to account for successful method transfer between different instruments. The Vanquish UHPLC system provides an entirely new and unique combination of active eluent pre-heating and advanced column thermostatting modes.

Separation techniques such as hydrophobic interaction chromatography, size exclusion chromatography, ion-exchange chromatography, and reversed-phase chromatography (RPC) are routinely applied to profile the therapeutic protein during this stage of development. RPC can be run with mass spectrometry (MS)-compatible mobile phase, hence the method can be easily transferred to MS characterization laboratories when required. In this work, the MAbPac-RP column was operated with the Vanquish Flex UHPLC system for the RPC of several intact therapeutic antibodies. To assess the suitability of the approach for stability studies, the separation of a reference and a stressed mAb chromatogram was compared.

WatchIs Retention Time Precision Only Affected by the LC Pump?
Retention time precision is extremely important in being able to accurately compare results. Traditionally, retention time precision was always attributed to flow characteristics, such as consistency in flow rate and gradient precision, in an LC system. However, with UHPLC a point has now been reached where other factors, such as pressure and speed, play major roles. Watch this video to learn which other factors are important as well as ways to generate very accurate retention time precision at the top of the UHPLC operating range.
